Anti-Ulcerative Potential of Egyptian Propolis against Oxidative Gastric Injury Induced by Indomethacin in Rats

چکیده
The anti-ulcerative and antioxidant potentials of Egyptian propolis alcoholic extract as well as its total phenolic and flavonoid contents were evaluated. The content of total phenols and flavonoids of propolis extract were determined by spectrophotometric methods with an FC reagent and by the complexation reaction with aluminium chloride, respectively. The antioxidant activity was examined with 1,1-diphenyl-2-picryl-hydrazyl (DPPH) and superoxide anion radicals scavenging assays. Indomethacininduced ulcer with or without pylorus ligation was used as the gastric ulcer model in rats. Oxidative stress was evaluated by measuring stomach homogenate levels of GSH, MDA and NO. The effects of the extract on gastric juice volume and acid output were evaluated. The total phenolic and flavonoid contents of propolis extract was 87.65 ± 0.5 mg GAE/g and 113.14 ± 0.86 mg QE/g extract respectively. The in vitro antioxidant activity was demonstrated by its ability to quench free radicals generated by DPPH and superoxide anion radicals, with the IC50 9.97 ± 0.45 and 11.29 ± 0.65 μg/ ml, respectively. Animals pretreated with propolis extract (200 and 400 mg/kg, b.w.) before indomethacin-induced ulcer model resulted in significant decrease in gastric ulcers and improved oxidative balance in gastric mucosal tissues. These findings indicate that Egyptian propolis possesses potent anti-ulcerative and antioxidant activities, corroborating the folk use of propolis preparations, and contributing for its pharmacological validation.

Background: Data suggest that V. arctostaphylos, B. thunbergii var. atropurpurea, E. angustifolia and L. acanthodes may prevent gastric ulcers induced by Non-steroidal anti-inflammatory drugs (NSAIDs). Objective: To explore the ulcer-protective potential of these plants in indomethacin-induced gastric ulcers in rats. Methods: Saline, hydro-alcoholic extract of each plant (100, 400, 800 mg/...
متن کاملGastroprotective effect of sodium hydrosulfide against indomethacin-induced gastric ulcer in diabetic rats
Introduction: The incidence rate of gastric erosions and ulcers in diabetic patients are higher due to failure of mucosal antioxidant defense and maintain enough blood flow. The present study evaluated the gastro-protective effect of sodium hydrosulfide (NaHS) against indomethacin-induced gastric lesions in diabetic rats. Methods: In order to test anti-ulcer activity of NaHS against indomet...

Background: Gastric ulcer is among the most serious stomach disorder universally. Several effective drugs are employed in the management of this disease, although there have been adverse effects in some cases. The aim of this study was to investigate the effect of nortriptyline to protect against gastric lesions, induced by indomethacin or cold-stress in rats. Methods: Gastric lesions were ind...

Background: Saffron is the dried stigmata of the flowers of saffron (Crocus sativus L., Iridaceae). Saffron is well known for the treatment of gastric disorders in traditional medicine. Objectives: In the search for new potential antiulcer agents, the effects of the ethanol extract of saffron and its active constituents crocin and safranal as compared with omeprazole against gastric ulcer i...
متن کاملThe anti-inflammatory and anti-apoptotic effects of gallic acid against mucosal inflammation- and erosions-induced by gastric ischemia-reperfusion in rats
The present study aimed to evaluate the protective effect of gallic acid on gastric mucosal lesions caused by ischemia-reperfusion (I/R) injury in rat. Forty male rats were randomly divided into sham, control (I/R injury) and three gallic acid-pretreated groups. To induce I/R lesions, the celiac artery was clamped for 30 min and then the clamp was removed to allow reperfusion for 6 hr. Pretreat...
